PCR——Classification of lab contamination

a. Cross-contamination between samples

It is mainly caused by cross-contamination between sampling tools in the process of sampling (taking), or improper use of pipettes, centrifuge tubes, etc. in the process of nucleic acid extraction.


b. PCR reagent contamination

It is mainly due to the contamination of pipettors, containers, negative controls and other reagents by nucleic acid template or positive control during the sample loading process of PCR plastic consumables.


c. PCR amplification product contamination

This is the main and most common contamination problem in PCR reactions. Because the PCR product is so large that it is much higher than the PCR detection limit of a few copies, contamination of very small amounts of PCR product can lead to false positives." Another easily overlooked form of contamination is aerosol pollution, which can cause PCR product contamination, and aerosol contamination is a problem that deserves special attention.


d. Contamination with cloned plasmids

Positive reference materials are often used in experimental operations. Most of these reference materials are made of some cloning plasmids. The concentration of plasmids in the unit volume is high, and it is easy to cause contamination if used carelessly.
